<ul>&#13; <li>ザクロ（<em>Punica granatum L.</em>）の未利用部位である葉?枝から1,2,3,4,6-Penta-O-galloyl-β-D-glucose（PGG）を同定しました。</li>&#13; <li>ザクロ葉?枝抽出エキスやPGGは、ATTRアミロイドーシスの原因であるTTRアミロイド線維を試験管内（<em>in vitro</em>）で分解することを見出しました。</li>&#13; </ul>&#13; <p>【概要説明】</p>&#13; <p>熊本大学大学院生命科学研究部附属グローバル天然物科学研究センター（薬学系）の首藤 剛 准教授、大学院生命科学研究部脳神経内科学講座の植田 光晴 教授、大学院薬学教育部博士後期課程の鏡 明日香 氏らの研究チームは、ザクロ（<em>Punica granatum L.</em>）の未利用部位である葉?枝から得られる天然物 PGG が、ATTRアミロイドーシスの原因となる トランスサイレチン（TTR）アミロイド線維を直接標的として分解することを見いだしました。熊本大学独自の天然物バンクを用いた植物スクリーニングから研究を開始し、試験管内（<em>in vitro</em>）-線虫内（<em>in vivo</em>）-患者検体（<em>ex vivo</em>）の複数の評価系で一貫した活性を確認しました。</p>&#13; <li>脂肪組織と肝臓の双方に作用し、APNとFGF21を同時に高める天然由来成分として、グネチンCは代謝性疾患に対する新たなアプローチの可能性を示しました。</li>&#13; </ul>&#13; <p>【概要説明】</p>&#13; <p>熊本大学大学院生命科学研究部附属グローバル天然物科学研究センター（薬学系）の首藤剛准教授および大学院薬学教育部博士課程の岸本朋樹氏らの研究チームは、株式会社山田養蜂場との共同研究として、インドネシア原産の植物「メリンジョ（Gnetumgnemon）」の種子から得られるポリフェノール「グネチンC」に、肥満および2 型糖尿病の改善作用があることをマウスモデルで明らかにしました。<br/>本研究では、高脂肪食で肥満状態となったマウスにグネチンC を4 週間投与し、体重、脂肪蓄積、空腹時血糖の改善効果を確認しました。その背景には、脂肪組織におけるアディポネクチンの高分子量多量体化を促すPPARγ–DsbA-L 経路の活性化、ならびに肝臓でのSirt1 依存的なFGF21 産生の増加が関わることを突き止めました。<br/>FGF21 は、肝臓から分泌され、脂肪細胞のβKlotho とFGFR1 を介してAPN 活性をさらに高める「正のフィードフォワードループ」を形成することも示唆され、これらの機序も関わり、結果として、APN とFGF21 を同時に活性化し、全身の代謝バランスが改善されることが示されました。<br/>この結果は、複数臓器を標的とする天然物治療の可能性を示す重要な成果です。本研究の成果は、Springer Nature の「Scientific Reports」（本年１１月２５日(日本時間)オンライン）に公開されました。</p>&#13; <p>インド大使館よりのメッセージが届けられ、日印の文化?学術交流の重要性が強調されました。参加者はラーマーヤナの物語や伝統衣装?料理を通じてディワリの多様な文化を学びました。</p>&#13; <p>最大の見どころは、色鮮やかな粉で模様を描く「ランゴリ」体験。創造性を発揮し、美しい模様が次々と完成しました。その後、サモサやチャイなどインドの伝統的なお菓子を味わいながら交流を深めました。</p>&#13; <p>熊本大学からは、経営企画本部国際事業戦略チームの職員名が参加し、大学の最新情報や特色を紹介するブースを設置しました。このブースには、中国の大学や公的機関、高校、日本語学校などから約<span>30</span>名が訪れ、学問分野や学科に関する情報や、入学条件について積極的な質問があり熊本大学への関心の高さがうかがえました。熊本大学は大学の特徴や魅力、熊本大学で学ぶことがいかに実り多いものであるかを伝えました。</p>&#13; <p>本シンポジウムは、大学や研究機関の歴史?文化資源の保存と活用をテーマに、台湾国内外の専門家や博物館関係者が集うもので、今回は日本から熊本大学の松永学長特別補佐、マレーシアからマラヤ大学の研究者が招かれ、台湾全土の大学?博物館関係者など<span>45</span>機関が参加しました。<span>2</span>日間で計<span>28</span>件の口頭発表と<span>4</span>件のポスター発表が行われ、活発な意見交換が展開されました。</p>&#13; <p>　松永学長特別補佐が「熊本大学キャンパスミュージアム：大学における芸術と文化の取り組み」と題して基調講演を行いました。講演では、熊本大学が誇る「第五高等学校」時代からの建築?文化財の保存や、教育?地域社会への文化発信の取り組みを紹介し、学術資産を次世代へつなぐ大学博物館の役割について述べました。</p>&#13; <p>　また、松永教授は「第五高等学校から現在の熊本大学へ」と題する写真展を同時開催し、熊本大学の歴史を伝える<span>24</span>枚の貴重な写真を展示しました。これらは来場者に大きな関心を呼び、講演内容とあわせて熊本大学の歴史と文化的価値を伝える貴重な機会となりました。展示資料は、今後の両大学の連携の象徴としてそのまま国立成功大学へ寄贈され、同大学で継続展示される予定です。</p>&#13; <p>　小川 久雄学長の挨拶に続き、「脳によるエネルギー代謝制御について」について、大学院生命科学研究部の 戸田 知得 准教授が説明しました。戸田准教授は、脳による代謝制御のメカニズムをテーマに研究を進めており、私たちが食事をとる前、すなわち「食べることを予測した段階」で、脳がすでに血糖値や食欲を調整しているという仕組みに注目し、その神経活動を解明することを目指していると説明しました。本研究は、<span>2024</span>年度「創発的研究支援事業」に採択されるなど、今後の発展が期待される先端的なテーマです。</p>&#13; <p>　次に、「セミコン台湾<span>2025 </span>学生交流プログラム」について、大学院自然科学教育部 情報電気工学専攻 修士２年の園田 康太郎さんと、大学院自然科学教育部 半導体?情報数理専攻 修士１年の程 識恩さんが参加報告を行いました。程さんらは半導体産業への関心や理解を深めることを目的とした台湾での企業訪問や展示会の見学、台湾大学生との交流を通し、貴重な経験や多くの学びを得ることができたと話しました。</p>&#13; <p>　次に、「第２回九州学生映画祭」について、工学部３年 園川 凌真さん（第２回九州学生映画祭実行委員長）が活動報告を行いました。園川さんは、?九州学生映画祭から羽ばたく未来の商業映画監督を輩出する。?という本映画祭における長期的な目標を話すとともに、今年度の<span>9</span>月に開催された第２回九州学生映画祭では、事前交流会や企業協賛?名義後援、行定 勲 監督のトークショーを実施したことを話しました。</p>&#13; <p><span>?　</span>次に、共創学環総合型選抜入試の志願倍率について、教育?学生支援担当理事 水元 豊文 理事が説明を行いました。水元理事は、本入試の志願倍率は実施発表当初の予想を大きく超えるものとなったことを説明すると共に、今後も多くの高校生や保護者へ情報が広がっていくことを願うと話しました。</p>&#13; <p><span>?　</span>最後に、物価高騰緊急支援寄附募集キャンペーンについて総務?財務?施設担当理事 黒沼 一郎 理事が説明を行いました。黒沼理事は、昨今の急激な物価高騰という厳しい社会情勢は本学の節減努力を超えており、各種経費の支出増加により、将来の学生のための資源さえも一部切り崩して対応せざるを得ない事態となっているため、今回緊急支援の寄付募集を行うこととしたと話しました。また、<span>11</span>月<span>12</span>日（水）から開始した寄附募集キャンペーンへ多くの方にご参加頂き、より良い教育環境の整備を目指したいと話しました。</p>&#13; <li>今後、量子情報処理や高性能量子メモリなどへの応用が期待されます。</li>&#13; </ul>&#13; <p><strong>【概要説明】</strong></p>&#13; <p>熊本大学大学院先端科学研究部の速水真也教授らの研究グループは、韓国?台湾の共同研究者とともに、コバルトイオンが金属<strong><span>-</span></strong>金属結合によりつながった錯体分子「<span>Co</span>?(dpa)?Cl?（<span>dpa</span>＝<span>2,2</span>′<span>-</span>ジピリジルアミド）」が、量子ビット（スピン量子ビット）^{※<span>1</span>}として機能することを世界で初めて明らかにしました。</p>&#13; <p>本研究では、パルス電子スピン共鳴（<span>EPR</span>）法^{※<span>2</span>}を用いてスピンの緩和特性を詳細に解析し、金属<strong><span>-</span></strong>金属結合をもつ分子でも比較的長いスピン寿命を保持できることを実験的に証明しました。</p>&#13; <p>この成果は、分子を基盤とした量子コンピュータ材料設計の新しい方向性を示すものであり、将来的には量子情報処理技術やスピンエレクトロニクスの発展に貢献することが期待されます。</p>&#13; <p>本研究成果は、ヒトのエネルギー代謝を担うミトコンドリアの仕組みの理解や、ミトコンドリア病^[4]などミトコンドリア翻訳異常に関わる疾患の理解につながるものと期待されます。</p>&#13; <p>ヒトの細胞の中には細胞質で起こる翻訳以外にもミトコンドリアの内部で起こる翻訳が存在します。ミトコンドリア内の翻訳を網羅的かつ高解像度に多検体で解析する手法は、望まれつつもこれまで存在しませんでした。</p>&#13; <p>今回、共同研究グループは、新たに「MitoIP-Thor-Ribo-Seq法^[5]」という手法を開発し、ミトコンドリア翻訳速度の計測や、ミトコンドリアtRNA（mt-tRNA）^[6]修飾による翻訳の促進効果、ミトコンドリア病患者の細胞での翻訳制御不全といった、複雑な翻訳動態を明らかにしました。</p>&#13; <p>　本研究は、科学雑誌『<em>Molecular Cell</em>』オンライン版（11月12日付：日本時間11月13日）に掲載されます。</p>&#13; <p/>&#13; <p><strong>（今後の期待）</strong></p>&#13; <p>MitoIP-Thor-Ribo-Seq法は、ミトコンドリア翻訳の動態を網羅的かつ高解像度に多検体で測定できる新たな技術です。今回明らかになったミトコンドリア翻訳速度の多様性やmt-tRNA修飾の重要性は、ミトコンドリア病や加齢に伴うエネルギー代謝の機能不全や、その細胞種?組織不均一性の理解につながると期待されます。また、本手法はミトコンドリア翻訳異常を発端とするさまざまな疾患の治療標的の探索、ミトコンドリア翻訳の活性化を目的とする創薬研究にも応用が見込まれます。今後、さまざまな細胞や組織に本手法を適用することで、ミトコンドリア翻訳の多様性や調節メカニズムを解き明かし、エネルギー代謝の理解を大きく前進させることが期待されます。</p>&#13; <p/>&#13; <p><strong>（補足説明）</strong></p>&#13; <p><sup/></p>&#13; <p><strong>[1] ミトコンドリア</strong></p>&#13; <p>真核細胞内に存在する小器官で、独自のDNAを持ち、アデノシン三リン酸（ATP）を産生する「エネルギー工場」。</p>&#13; <p><strong>[2] 翻訳</strong></p>&#13; <p>メッセンジャーRNA（mRNA）（[7]参照）に記された塩基配列をアミノ酸配列へ変換し、リボソーム（[8]参照）でアミノ酸を結合してタンパク質を合成する過程。</p>&#13; <p><strong>[3] ミトコンドリア翻訳</strong></p>&#13; <p>ミトコンドリアDNAから写しとられたmRNAからタンパク質を合成する過程。ミトコンドリア内で、専用のミトコンドリアリボソームによって行われる。</p>&#13; <p>ミトコンドリア免疫沈降法（MitoIP）とRNA増幅法（Thor（[11]参照））を組み合わせることでミトコンドリアリボソームの解析に特化したRibo-Seq法（[9]参照）。網羅的かつ高精度にミトコンドリア翻訳を解析できる。</p>&#13; <p><strong>?[6] ミトコンドリアtRNA（mt-tRNA）</strong></p>&#13; <p>ミトコンドリアDNAにコードされる22種類の転移RNA（tRNA）の総称。ミトコンドリアリボソームにmRNA情報に基づいたアミノ酸を供給し、翻訳伸長（[14]参照）を支える。</p>&#13; <p><strong>[7] メッセンジャーRNA（mRNA）</strong></p>&#13; <p>タンパク質のアミノ酸配列情報（コドン）を持つRNA。リボソームによってコドンが読み取られることでタンパク質が合成される。</p>&#13; <p><strong>[8] リボソーム</strong></p>&#13; <p>リボソームRNA（rRNA）とリボソームタンパク質で構成される巨大複合体。</p>&#13; <p><strong>[9] Ribo-Seq法</strong></p>&#13; <p>翻訳装置であるリボソームを抽出し、リボソームと結合しているmRNA配列を同定することで、どの遺伝子がどの程度の効率で翻訳されているかを解析する手法。リボソームは大きな複合体であり、一定のmRNA領域を覆うように結合するため、これらのリボソームとmRNAの複合体をRNA分解酵素で処理すると、リボソームが保護するmRNA断片だけが分解されずに回収される。</p>&#13; <p><strong>?[10] 免疫沈降法</strong></p>&#13; <p>特定のタンパク質やタンパク質複合体を抗体によって沈殿させ、目的の分子や小器官を選択的に回収?濃縮する方法。</p>&#13; <p><strong>?[11] Thor</strong><sup/></p>&#13; <p>T7 RNAポリメラーゼを用いて、RNAを鋳型にRNAを増幅する手法。Ribo-Seq法において、フットプリントの増幅に用いられる。ThorはT7 high-resolution original RNAの略。</p>&#13; <p><strong>?[12] ミトコンドリアリボソームランオフアッセイ法</strong></p>&#13; <p>一定時間を置きmRNA上で翻訳反応（ラン／run）しているミトコンドリアリボソームに、翻訳を終結させ、乖離（かいり）（オフ／off）させる実験手法。ミトコンドリア翻訳開始を阻害することで実験が可能になる。</p>&#13; <p><strong>?[13] Ribo-Calibration法</strong></p>&#13; <p>　他方で、昨今の物価高騰は、これまでの節減努力等を超えており、業務経費の増加の中で質を切り下げずに教育研究?サービスを継続していくために、施設設備の修繕費用など将来の学生のための資源をも切り崩して対応している状況もあります。</p>&#13; <p>　このような中で、大学が前向きに発展を続けていくため、本学執行部としては、これまで以上の業務効率化を図ることは当然として、皆様方から寄附等のご協力をお願いできないかと考えております。</p>&#13; <p>　In response to the rapid social changes surrounding universities, including transformations in regional and industrial structures such as the semiconductor-related industries, our university has been proactively making new investments to strengthen educational and research activities. These efforts include the establishment of new undergraduate programs and schools and graduate courses to meet emerging needs for human resource development and students’ aspirations for higher education. At the same time, the university has pursued financial reforms such as securing self-generated revenue and reducing expenses. Through these initiatives, we have been recognized as a “Core Research University in the Region”, and have become one of the national universities attracting significant attention nationwide as our region continues to evolve.</p>&#13; <p>　However, the recent surge in prices has gone far beyond what can be absorbed through conventional cost-saving efforts. To maintain the quality of education, research, and services despite rising operational costs, we have, in some cases, been compelled to draw upon resources originally reserved for future students, such as funds for facility repairs and renewal.</p>&#13; <p>　In this context, to ensure the university's continued positive development, the university administration recognizes the need not only to further enhance operational efficiency, but also to seek broader cooperation from the university community, including through donations and other forms of support.</p>&#13; <p>　In making this request for cooperation, we would like to take this opportunity to discuss with you regarding the future direction of our educational and research environment, including updates to facilities, equipment, and systems, together with an overview of the university’s financial outlook.</p>&#13; <p>　Our university remains firmly committed to contributing to both local and international</p>&#13; <p>communities as an “educational hub university open to the region and the world, contributing to society through co-creation.” We will continue striving to maintain and enhance our educational and research environment.
